Fluorescence measurement is a photometric measurement principle based on the detection of light radiation generated by fluorescence. The excitation light shines through the sample, which contains the fluorescent substance. This transforms part of the incident light of wavelength λ into fluorescent light of a longer wavelength λ1 The light thus produced radiates in all directions and can be measured with a detector, e.g. perpendicular to the axis of the incident light. The radiant power of the fluorescent light is proportional to the concentration of the fluorescent substance in the range of small amounts. The main reasons for using fluorescence measurement are higher selectivity as well as greater sensitivity compared to absorption measurement.
